چکیده
Abstract Background Activation of autophagy flux contributed to resistance breast cancer (BC) cells current chemotherapeutic drugs, which seriously limited their therapeutic efficacy and facilitated BC recurrence in clinic. However, the detailed mechanisms are still not fully understood. In present study, we identified that inactivation AMPK-ULK1 signaling cascade mediated protective sensitized doxorubicin vitro. Methods Cell counting kit-8 (CCK-8) assay colony formation were performed evaluate cell proliferation abilities. Trypan blue staining was used examine viability, Annexin V-FITC/PI double method conducted determine apoptosis. The autophagosomes observed photographed by electronic microscope (EM). Western Blot analysis employed genes expressions at protein levels. Results parental doxorubicin-sensitive (DS-BC) exposed increasing concentrations establish doxorubicin-resistant (DR-BC) cells, DR-BC much more resistant high-dose treatment compared DS-BC cells. Interestingly, specifically increased LC3B-II/I ratio, promoted decreased p62 expression levels facilitate instead inhibitor 3-methyladenine (3-MA) enhanced cytotoxic effects on addition, proved triggered activating pathway. Functionally, phosphorylated AMPK (p-AMPK) ULK1 (p-ULK1) activate pathway inhibitors for (compound C) (SBI-0206965) blocked promote death slow down growth treated with doxorubicin. Conclusions Collectively, our vitro data indicated blockage might be a promising strategy increase sensitivity treatment.

15 صفحه اولAMPK-mediated autophagy inhibits apoptosis in cisplatin-treated tumour cells
The role of autophagy in cisplatin anticancer action was investigated using human U251 glioma, rat C6 glioma and mouse L929 fibrosarcoma cell lines. A dose- and time-dependent induction of autophagy was observed in tumour cells following cisplatin treatment, as demonstrated by up-regulation of autophagy-inducing protein beclin-1 and subsequent appearance of acridine orange-stained acidic autoph...
متن کاملActivation of the AMPK-ULK1 pathway plays an important role in autophagy during prion infection
AMPK is a serine/threonine protein kinase that acts as a positive regulator of autophagy, by phosphorylating ULK1 at specific sites. A previous study demonstrated activation of the macroautophagic system in scrapie-infected experimental rodents and in certain human prion diseases, in which the essential negative regulator mTOR is severely inhibited. In this study, AMPK and ULK1 in the brains of...
متن کاملAMPK activates autophagy by phosphorylating ULK1.
In eukaryotes, macroautophagy (hereafter, autophagy) functions as a highly conserved catabolic mechanism in which cytoplasm, including damaged organelles and protein aggregates, is sequestered into autophagosomes, double-membrane vesicles, for ultimate degradation and recycling in lysosomes. Autophagy has attracted increasing attention in recent years because of its significance in various aspe...
